CLEC12A属C型凝集素受体家族，广泛参与免疫稳态调控。尽管该家族成员功能多样，CLEC12A在恶性肿瘤中的预后价值及其在肿瘤免疫微环境中的作用尚未系统阐明。通过整合TCGA、GTEx、HPA等多组学数据库资源，联合GSEA富集分析及R语言生态工具，系统评估了CLEC12A在33种实体瘤中的表达特征、预后意义及其与免疫浸润的关联。结果表明：CLEC12A在22种肿瘤组织中呈异常高表达或低表达，且其表达水平与多癌种患者总生存期显著相关，关联方向呈现癌种特异性。功能富集分析提示，CLEC12A显著富集于调控T细胞活化、髓系细胞分化及细胞因子-细胞因子受体相互作用等免疫相关信号轴。免疫浸润层面，CLEC12A表达与CD8~+T细胞、M1/M2型巨噬细胞、树突状细胞及中性粒细胞等多类免疫细胞浸润水平显著相关。综上，CLEC12A可作为多种恶性肿瘤的独立预后标志物，并具备成为免疫治疗潜在靶点的转化价值。
Abstract：CLEC12A is a member of the C-type lectin receptor family and critically maintains immune homeostasis. Although extensive functional heterogeneity has been described for other family members,the prognostic significance of CLEC12A and its role in shaping the tumor immune microenvironment(TIME) remain poorly characterized across human malignancies. Here,we leveraged multi-omic data from TCGA,GTEx,and HPA repositories,integrated gene-set enrichment analysis(GSEA) and an R-based bioinformatic pipeline,and systematically interrogated CLEC12A expression profiles,prognostic value,and immune-infiltrative features across 33 solid tumor types. CLEC12A was aberrantly up-or down-regulated in 22 tumor types compared with matched normal tissues,and its expression level showed cancer-specific correlations with overall survival of patients. GSEA revealed significant enrichment of CLEC12A in immune-related signaling axes governing T-cell activation,myeloid cell differentiation,and cytokine–cytokine receptor interactions. Immune-deconvolution analyses further demonstrated that CLEC12A expression tightly correlates with intratumoral abundances of CD8~+T cells,M1/M2 macrophages,dendritic cells,and neutrophils. Collectively,CLEC12A constitutes an independent prognostic biomarker and represents a therapeutically exploitable immune target in multiple human cancers.
