瓜蒌皮抗肿瘤活性成分的初步研究

摘要:

采用改进的MTS法来探究瓜蒌皮石油醚相不同极性成分对结肠癌HCT-116细胞和乳腺癌MCF-7细胞增殖的抑制作用,探讨其抗肿瘤活性。利用硅胶柱色谱制备极性不同的五种成分,改进的MTS法检测细胞的增殖作用,Propidium Iodide染色法分析HCT-116细胞周期;Annexin V-FITC/Propidium Iodide染色法分析HCT-116细胞凋亡。在低浓度情况下发现,瓜蒌皮醚相极性成分促进MCF-7细胞生长,成分E诱导细胞停滞在S-期而起到抑制癌细胞增殖的作用,并且成分C及E抗癌细胞增殖作用是通过诱导细胞凋亡介导的作用机制,并以剂量依赖方式诱导其细胞凋亡。通过硅胶柱色谱制备的石油醚相不同极性成分具有抗肿瘤活性。

Abstract:

Improved MTS method was used to explore the inhibitory effect of different polar components in human colon cancer HCT-116 cells and breast cancer MCF-7 cell proliferation gualoupi petroleum ether,and its antitumor activity was explored. Using silica gel column chromatography to prepare five different components with different polarity,the proliferation of MTS cells was detected. The improved method Propidium Iodide staining was used for the analysis of HCT-116 cell cycle. Analysis of apoptosis of HCT-116 cells was by Annexin V-FITC/Propidium Iodide staining. In the case of low concentration,gualoupi ether polar components promote the growth of MCF-7 cells. E components induce the cell cycle in S-phase to inhibit the proliferation of cancer cells; E cancer cell proliferation is through the mechanism of inducing apoptosis mediated,and inducing apoptosis in a dose dependent manner. The petroleum ether phase with different polar components prepared by silica gel column chromatography has antitumor activity.
